MauriceFlex™ for protein fractionation

Presenting MauriceFlex, a novel protein fractionation system that enables users to collect fractions for the analysis of charge variants. Users of MauriceFlex receive:

  • The ability to work with fractions more freely: analyze with intact mass, peptide mapping, reduced and subunit masses, and more.
  • Release from arduous high-performance liquid chromatography (HPLC) and ion-exchange chromatography (IEX) procedures: readily collect high-purity fractions without the need for further, time-consuming chromatography-based fractionation development.
  • The ability to accomplish anything on one’s own: imaged-capillary isoelectric focusing (icIEF), CE-SDS, and icIEF protein fractionation all in one instrument.
  • The ability to use time more freely: charge variations can be separated, fractionated, and identified in less than a day.

The MauriceFlex integrates icIEF, CE-SDS analysis, and protein charge isoform fractionation into one unit, enabling users to go beyond routine charge and size and perform charge variant analysis of their molecule at any point without requiring the use of separate techniques like IEX and HPLC.

Using IEX or HPLC for protein fractionation?

Compared to IEX, HPLC, and other existing technologies, protein fractionation with MauriceFlex yields considerable time and capital savings. The principal benefits are:

  • Same-day fraction collection
  • High purity fractions
  • Compatibility with methods containing methylcellulose and urea
  • One fractionation run providing sufficient material for intact and reduced mass analysis
  • Ability to pool fractions from multiple runs for peptide mapping
  • The same system offering CE-SDS and cIEF assays

How protein fractionation works on MauriceFlex

  1. Exposure to an electrical field produces a pH gradient across the capillary within the MauriceFlex cartridge.
  2. Proteins separate by moving to different pH values on the gradient, based on their unique isoelectric point (pI).
  3. When ammonium acetate is added, the pH gradient is altered, which mobilizes the divided proteins and causes elution (fractionation).
